Buffalo spermatozoa are more susceptible to cryodamage than cattle with poor post-thaw motility and fertilization failure. Cryopreservation leads to osmotic stress/mechanical stress to sperm leads to poor freezability and post thaw semen quality parameters. There are different methods or techniques developed to minimize the cryocapacitation, reduction of oxidative stress, improvement of post thaw sperm viability and fertility. One such method is exposure of exogenous cholesterol prior to processing. In many species, exogenous cholesterol (CLC) has increased semen quality parameters and in-vitro fertility. The CLC protects the sperm from cryoinjury by different mechanisms. CLC improves the plasma membrane and mitochondrial stability of spermatozoa, minimizes oxidative stress & leakage of intracellular enzymes and improves fertility rate. CLC minimizes the membrane fluidity and enhances organization or stabilization of membrane. CLC technology enhances the number of bulls as the semen donors. This book or monograph will be very useful to the scientists working in andrology laboratory, frozen semen bank & artificial insemination centres, animal breeders and buffalo farmers.
